  • 药理作用:丙酸睾丸素又称丙酸睾酮、丙酸睾丸酮,常温下为白色结晶或类白色结晶性粉末,无臭。不溶于水,易溶乙醇或乙醚,略溶于植物油,极易溶于氯仿。药理作用与甲睾酮相似,但作用较后者略强。因口服易被肝脏破坏,故口服无效。常肌注其油溶液,吸收缓慢,作用强而持久,可维持时间数日。丙酸睾丸素能促进男性器官及第二性征的发育,促进蛋白质合成及骨质形成,对抗雌激素,抑制子宫内膜生长及卵巢、垂体功能。用于无睾症、隐睾症、男性性腺机能减退症、妇科疾病如月经过多、子宫肌瘤、老年性骨质疏松及再生障碍性贫血等。 本品在体内先转化为5α-二氢睾酮 (5α-dihyrotesterone),以后再与细胞受体结合,进入细胞核,与染色质作用,激活RNA多聚酶,促进蛋白质合成和细胞代谢。此外,丙酸睾酮可通过红细胞生成素刺激红细胞的生成和分化。丙酸睾丸素口服虽可吸收,但在肝中会迅速破坏而失效,故一般采用肌内注射。肌内注射丙酸睾酮后,吸收较慢,其延效时间2~4日。在血中,98%的睾酮与性激素球蛋白结合,仅2%为游离状。半衰期为10~20min。睾酮在肝内灭活后,代谢产物为雄酮、异雄酮及原胆烷醇酮。它们90%是与葡萄醛酸及硫酸结合的形式从尿中排出。约6%非结合形式的产物是由胆汁排出,其中少部分仍可再吸收,形成肠肝循环。
