  • 酪氨酸激酶:酪氨酸激酶也叫酪氨酸蛋白激酶,是催化肽链中酪氨酸残基(位点)磷酸化的酶,在生物代谢和细胞信息传递中起重要作用,其分子量为56000,系细胞内膜结合的酶。该酶由两个结构域组成,一个是催化结构域,占酶分子的大部分,其中包含C端和两个酪氨酸磷酸化位点;另一个是调节结构域(即SH2结构域),包含豆蔻酰化的N端、丝氨酸磷酸化位点及同源区。 催化结构域中的酪氨酸磷酸化位点与酪氨酸激酶活性的表现关系很大,如果这些酪氨酸位点被磷酸化,激酶活性就会受到抑制;相反,若这些磷酸化酪氨酸位点用磷酸酶处理,使其去磷酸,则激酶活性可明显增加。酪氨酸磷酸化位点靠近C端处,但不同基因表达的酪氨酸激酶(如c-src基因家族的产物),这些位点所处位置不尽相同,如果将其缺失掉,激酶就被激活。此外,多瘤病毒中的T抗原与c-src基因表达的酪氨酸激酶结合的部位在邻近527位酪氨酸位点处,由于离这个位点太近,形成了空间位阻,妨碍了该位点的磷酸化,故增加了激酶的活性。 SH2结构域除同源区外,其他部分在结构上差异较大,因此可以与不同的调节因子作用,起到调节激酶活性的作用。
