  • 方法一:由苯酐经胺化、降解、酯化、重氮化、置换、氯化、环合、酸析、中和等步骤而得。 (1)邻氨基苯甲酸甲酯的制备。将苯酐和0℃的氨水依次加入反应釜,升温至50℃后缓慢滴加氢氧化钠溶液,保持在温度低于70℃和PH8.5-8.9条件下胺化,然后调PH=12-13,在65-70℃保温0.5h降解,完毕后保温赶氨3.5h。冷却至-10℃,加入-10℃的甲醇和次氯酸钠溶液(勿过量),在0℃下酯化45min,再逐渐升温至30℃,以淀粉碘化钾试纸测试呈无色反应。然后加入适量20%的亚硫酸氢钠溶液,料液转稀后,升温至50℃以上,再加入80℃的热水搅拌溶解,静置后过滤,分取油层得邻氨基苯甲酸甲酯。 (2)邻磺酰氯苯甲酸甲酯的制备。先将混酸置于重氮锅内,在10℃左右开始滴加邻氨基苯甲酸甲酯和亚硝酸钠溶液的混合液,重氮化温度保持在25℃以下,反应终点时淀粉碘化钾溶液显淡绿色。重氮化完毕后降温至10℃,加入硫酸铜,溶解后通入SO2进行置换,此时析出邻亚磺酸苯甲酸甲酯,约1h后用H酸试纸监测反应终点应显无色。然后加入甲苯,于30-35℃通氯气氯化,以2%联苯胺乙醇溶液测试显深墨绿色为终点。静置分层,有机层为邻磺酰氯苯甲酸甲酯甲苯溶液。 (3)不溶性糖精的制备。依次将水和邻磺酰氯苯甲酸甲酯甲苯溶液加入反应锅,在10℃时加氨水,搅拌反应15min(温度可达70℃,PH9以上),用2%联苯胺吡啶溶液测试不立即显黄色为环合终点。静置后取下层铵盐液,加入甲苯和30%的盐酸,降PH至1以下。酸析后降温至20℃,取甲苯层水洗去氯化铵得糖精甲苯溶液。 (4)可溶性糖精的制备。将不溶性糖精甲苯溶液加热至40℃,加入碳酸氢钠和水调PH至3.8-4,静置后取水层,加活性炭脱色、过滤,调滤液PH至7,再加活性炭脱色一次。滤液在70-75℃减压浓缩,保持PH为7趁热过滤。滤液经冷却、结晶、甩滤、干燥得糖精钠。
