  • 方法一:三甲基乙酰氯的制备有酸的直接氯化、叔丁基氯与一氧化碳在催化剂存在下直接合成或用异丁醇与甲酸反应生成三甲基乙酸,再与三氯化磷反应等方法,但大多数情况下采用第一种方法。由于三甲基乙酰氯的质量对合成药物的质量和收率影响很大,因此酸的氯化工艺尤其是选择恰当的氯化剂很关键。国外专利大都选用光气、苯甲酰氯作氯化剂,国内曾有人用过三氯化磷。河北轻化工学院通过实验发现,能符合药物合成要求的三甲基乙酰氯,可以在己内酰胺催化下以氯化亚砜作氯化剂制取,收率可达96%以上,含量达99%以上。 氯化亚砜氯化法的反应方程式如下:(CH 3 ) 3 CCOOH+SOCl 2 →(CH 3 ) 3 CCOCl+SO 2 ↑+HCl↑ 向装有电动搅拌器、温度计、滴液漏斗及连有导气管冷凝器的四口烧瓶中加入306g(3mol)三甲基乙酸和1.5g催化剂己内酰胺,加热熔化后搅拌,滴加氯化亚砜396g(3.3mol),保温缓缓搅拌至几乎无气体逸出为止。加热至70℃,减压蒸馏,分馏出氯化亚砜后再进行常压精馏,收集105℃馏分得产品354g,产率98%。三甲基乙酸的合成反应如下:(CH 3 ) 2 CHCH 2 OH+HCOOH[H 2 SO 4 ]→(CH 3 ) 3 CCOOH
